I have installed AEM Tuner 0.97d.
It works, it shows calibrations, but when I tried to connect to the ECU, I got USB Error, no matter does the ECU connected or not.
I use USB to COM device, it works with AEM Pro.

Go to ECU -> ECU Connection Preference and check "Serial first", then connect, do you still get the error?
BTW, is GEN 1 or GEN 2?

If I choose "Serial First" and try to connect, then the error occurs after 1-2 seconds, if I choose  "USB First" then the error occurs immediately.
Sorry, what is BTW??

Sorry I didn't make it clear. I was wondering to ask is your EMS the new "Series 2 " or the old one?

About the error:
AEMTuner will try to connect to EMS through serial port(COM) or USB. If you chose "Serial First", AEMTuner will look for EMS on com ports, if not found, then it will check usb ports.
In your case, AEMTuner cannot find EMS neither on serial or usb.
